Clipping layers: setting a layer to clip will make it paint over the color blob on the layer below. This is great for creating colored lineart, not drawing out of the lines, not having to reselect everything if you have to save and come back to the artwork later, great for color editing, and great for shading.
Multiply and Overlay layers: This is a layer setting that will make the layer you change to one of these have a specific effect. Multiply it will turn the layer into a transparent darker luminescent which is really great for shading and shadows. Overlay is the exact opposite where it creates a luminescent transparent layer that looks like light.
Folders: in some art programs like klips studio paint and sai You can use the above effects on the folders themselves making organization even easier. Especially since you won't need to set each individual layer if you want all the layers to be say a clipping layer.
Shadows: bonus a great way to create really good shadows easily is to fill the entire layer with the shadow color and then erase where the light hits. Brain seems to understand better where the light hits then how to place a shadow.
