I do. openCanvas has this weird glitch that makes it nearly impossible to completely remove any marks you make, even after erasing. There's always a very faint ghost of the line, and the only real way to remove it is lasso tool + delete button. D: It's just easier to make a new layer to ink and then get rid of the sketch layer.
My friend does. He makes a different layer for EVERYTHING @_@
Personally, the scanner I use is good enough to get nice blacks when I want to so I've never touched the ink outlines on a pic I'm about to digitally color. But I'm a terrible person to look at because I tend to do everything in one layer when I'm not specifically trying to spread things out (with someone over my shoulder telling me to) XD
Whenever I actually do ink (I should do it more... ) Yes, always a new layer, unless I managed to ink it traditionally, whoa, I can still ink traditional? heh heh

Unless there's something special about the brush. Then I work on a copy of the main layer.
And unless the result doesn't have to be clean. (90% of cases)

Cept for a few that I manage by inking freehand, but those are usually small doodles.
