Question for story-based Writers/Comicers

You have entered Da Blitz Arena... :D What it do folks?
As you may remember, I plan on a starting a story-oriented comic centered in BW2. I've done a few short comics here and there that were story-bound, but not only was that at least a year ago, they didn't involve so much material. So I was wondering, my story-making friends, and I know there are quite a few of ya:
Before you start on a story, what aspects of the story do you try to have completed & which do you focus on after you've already begun?
By aspects, I mean anything from choosing protagonists to support characters to locations, etc. Even as simple as the dialogue and forked-roads. Right now, I'm lining up possible characters, but it feels like I'm missing something else before I can make an attempt to start.
Any and all advice would be greatly appropriated.

This is how we built the story for renegade using this method.
Might I suggest creating your own story and universe? By doing a fan comic you are bound by the rules of an existing universe. Just a thought.
And yes, while I understand that there are a few rules I'd have to stick by when making this, but as I mentioned b4:
Let naught, the rules of one world restrict the possibilities within it, nor your imagination!! :D
'sides that, this has been done many times before, I do consider this a stepping stone bettering my artistic talents and story telling.
