First you want to rip the audio. The easiest way to do this is to use a service that extracts the audio from a youtube file and creates an MP3 from it, like this tutorial describes: http://www.ehow.com/how_5401257_ext.....be-videos.html
This, unfortunately, generates a larger file for longer videos, so it may be impractical. Still, it's probably the easiest way to get the sound from it without much loss in quality. If this is too much of an issue, you may be able to use Audacity (see below) to record the sound from the video directly, but this can be a bit difficult to set up reliably and depends on what kind of sound card you have.

Second, you can use a program like Audacity to edit out the audio you don't want. Audacity is available here: http://audacity.sourceforge.net/
