Do you know how to make cartoon animation like "Adventure Time" and "Regular Show"?
What software do they use?
And, how much knowledge and skills of animation need for making animation?

It only takes basic knowledge to animate.
Once you know the "twelve principles of animation", the rest is pretty easy.
Toon Boom is expensive, isn't it?
there is a guy in youtube who is an independent animator with awesome skill who has a how to tutorial series that might help you get started
Harry Partridge, he created a show on youtube called starbarians , really good animation & graphics for a 1 man project
he does a tutorial series on HuHaTwo or HuHa 2! youtube channel called:

Animating is pretty easy if you can picture what an object or character would look like moving around, and especially if you can see the previous frame (like a light-box tracer) while making the next one. Then it's really easy to make a frame depicting accurate movement.
It's also easier if you repeat frames for looping sequences like running , spinning, etc.
Also, taking the same frame and editing a small part to make a minor movement.
You can easily make an animation in Adobe ImageReady CS, but it requires changing the opacity of each previous layer (frame). But it works perfectly!
