It's late, I can't sleep. Fan art time.
For some reason I just can't let go of the sketch because inking stuff loses so much in the transition, but it looks so much more "professional" ... whatevs it's 4am.
OpenCanvas for the sketch, MangaStudio for the ink...

In all seriousness I can't figure out how to use a majority of the options - but you don't technically "need" them to get to drawing. I'm assuming this is a lot like Photoshop and similar programs, where you have tons of options, but on any given day only use about 5% of a given program.
The inking - Kind of what the program is all about. Lots of options, brush customization, line correction (for those of us whom have a terminal case of 'butterpaws') and layers.
the interface - Tons of crap you probably won't use. the program is focused for the private doujinshi creator. All that said, it's fairly intuitive, taking a lot of homage from Photoshop and such programs. Brush switching can be a bit annoying, but I've heard you can save your custom brush settings at any given time...
It can output to photoshop, which is great.... but I haven't found any options to save to jpg or anything else other than its odd storybook format
I got the Debut version, which I think was about $50... I can't say I'm disappointed with the purchase. I despise inking and this is way easier and faster than I've been able to ink with photoshop or illustrator.

I have the professional version for my webcomic. My opinion was that it has a huge learning curve, and it's tendency to want to make layers if you so much as look crosseyed at it is a little weird, but really I love the program and probably couldn't do without it now.
