Depends on the pen really. For this I used a brush pen, which tends to run if I paint over it, so I did it last. Usually if it's just plain micron I will ink first, but they've changed their formula and it's more prone to running, so lately I've been inking last and just using my pencil marks as guides for the paint. Hope that helps!
