The show revolves solely around the Eds, the other cul-de-sac children, and the Kanker sisters. Subtle hints throughout the series pointed to where the cartoon took place, as it was revealed that the cul-de-sac is in a town called Peach Creek. As similar to the Peanuts series, no adults are seen, as well as the seeming lack of any other inhabitants and any other possible relations; besides Sarah being Ed's little sister and a the running gag of Rolf and his mysterious family relations

May, Marie, and Lee are three sisters who serve as the antagonists towards the cul-de-sac and its inhabitants. Lee is the leader with curly red hair that covers her eyes, Marie has blue hair, and May is the buck-toothed blonde. They live in the nearby trailer park called Park 'n' Flush. If there is one thing that all of the kids in the cul-de-sac can agree on, it's that they all hate the Kanker Sisters. While all the Kankers like the Eds, it has been shown that May prefers Ed, Marie for Edd, and Lee for Eddy. May is voiced by Erin Fitzgerald, except Season 3, when she was voiced by Jenn Forgie, Marie is voiced by Kathleen Barr, and Lee is voiced by Janyse Jaud.

Buttercup (voiced by E. G. Daily) is the toughest of the three. Her personality ingredient is spice, her signature color is green, and she has short black hair in a flip. She is a tomboy who loves to get dirty, fights hard and plays rough; she does not plan and is all action. Buttercup is the only Powerpuff Girl without a unique super power (aside from being able to curl her tongue as shown in the episode "Nuthin' Special"). McCracken originally wanted to name the character "Bud" until a friend suggested the name Buttercup
