the base line, or like....the outline of the character has a thicker pen stroke (size?) when compared to the detailing pen strokes like the definitions of the muscles, folds, etc

Oooh, I gotcha.
Well, when I draw, I always draw everything very thin. After the character has been drawn, I go back over the image and add depth to the lines, to help bring out depth.

So what size do you use on (SAI? O.o) for the brush strokes and canvas size.

I think canvas size plays a part in the line weight as well. More visual actual

They are never below 2000 x 2000, unless I am working on something that needs to be smaller. I'm constantly changing the brush size from small, medium, to large.
