Hrrrrrrrrrrrrm tough question.
I think it depends mainly on you target audience~
-If you want a whole stack to take to a convention to sell off, get a local printer and talk with them first about specifications and deals you can get with them before making an image~
-if you just want to have a design or more to have avaliabe to everyone online, i have heard good things about the RedBubble website. the prices are very resonable for buyers, shippng included~ and there's no limit on colours used i have personally handled one of their products and the quality is great too :'3 it will last~
Doing it the website way will drastially lower your cut of each shirt, but the initial costs wont be as high either.. when getting them printed witha firm it'll cost more, but the gain will be higher to~ it's hard to determine which is 'best' as i've only ever gone with the local option~ :'P
