Depends on their overall potential. In Twilight's case she'll constantly feel exhausted as long as she maintains it, like a workout that doesn't end. Too long and it could be dangerous. Its even more exhausting when she transforms multiple ponies from one end of the spectrum to the other. These two were probably a few tons overweight before Pinkie asked her to flip them over AND make them macro.
