Someone was basically experimenting with turning humans into werewolves, using I think the original Man-Wolf (the son of the newpaper editor who's always harassing Spider-man) as a lab animal. Cap simply ended up temporarily injected with the formula and wolfed out like many of the test subjects but being Cap remained in control enough of himself to still beat the bad guy and he was cured right afterwords.
