I've got this theory that the Medic from Team Fortress 2 is really a grown-up male version of the Little Sister: see, years ago Dr. Tananbaum was trying to create a "Little Brother", but his self-healing abilities were too slow-acting, so Andrew Ryan had him shipped out to the real world to be forgotten.
There, the Little Brother grew up, got his medical degree, and invented the Medigun, fueling it with the ADAM he naturally produced. Incidentally, notice how whenever a Medic is on a team, everyone demands to have his healing abilities- very much like the Splicers and the Little Sister. Not only that, but he has quite an affinity to hang around huge, hulking, dim-witted brutes who carry heavy weapons.
