That One Boss is that specific kind of boss in a game that fills you with dread every time you play the game and come accross him/her/it. In my case, That One Boss is the Nasty Metallic Enforcer (NME) that you see in the picture. It's from Apogee's Rise of the Triad FPS circa 1994 and is one of the bosses in the game.
So what's so bad about it? For starters, it moved damn fast and is capable of launching a number of heat-seeking missiles and mines that would hunt you down without fail. Not only that, when damaged enough it would spew out a wall of fireballs on occassion, guaranteed to kill you within seconds. Oh, and it would try and run you over as well.
To make matters worse, the only source of health was a locked room which you had to get a key for: a key located directly behind the spot where the NME is when it first sees you. Also, in that same room was a pair of patrolling robots called Ballistikraft that fired mines and were INDESTRUCTABLE. And heaven help you if the NME followed you into that room as well.
It was definitely the most difficult boss I've ever encountered when I played ROTT back in my early PC days, as it kept killing and was even more difficult than the final boss, El Oscuro. Even the developers started in the manual that "Even we have difficulty beating this guy". No joke. To this day, it is still the most memorable boss battle for me, especially since it included one of the best boss battle songs I've ever heard.
Recently, I got ROTT running on DosBox and came to the point where I faced the NME. This is 14 years later, when I've learned many FPS tricks and methods during that period. I went in for the showdown...

"Silly player! Just because you've learned new tricks doesn't mean I can't nuke you on the spot."
