I dunno how this meeting happens, but supposing it does and it’s because of unethical magics performed by vamponies, and Twily gets involved. The backstory I got out of Blood Moon here, is that Moon is the safest magical expert of the vamponies for a mortal or Sun-aligned pony like Twi to talk to.

But yeah, instances of certain powerful vamponies openly deploying spells that pose significant threat to the safety of the realm and its denizens, so Twilight gets to go investigate it. She gets pinballed around a bit before she meets Blood Moon, and it gets heated during discussions. Moon is not fond of helping anyone besides herself and the vamponies of the Sanctuary. So she starts making digs at Twi to make her leave. But we all know Miss Sparkle and her doggedness to do right. This picture is one of those moments in the discussions. c:
