I remember we previously saw the poor wolf with his unfortunate rat phobia before when he found himself in a duel with Eternal, so this definitely would be a hellish experience for him.
It looks like this is the moment when he'd first become aware of the rat, perhaps feeling the brush of whiskers against his foot as he hastily pulls it away. The rat having been curious about this new visitor and being rather friendly and inquisitive might think that the wolf is playing. Perhaps a game will ensue of the rat attempting to jump onto Don Lope's foot as he tries to keep on moving his feet around to avoid contact. His muffled and terrified murmurs might sound rather like squeaking to the rat whom might naturally think that the wolf is enjoying the game as much as he is.
Sadly for Don Lope, the rat might have decided he's a new friend and be determined to keep company with him while being very playful and affectionate.

Another thought is that perhaps the rat is attempting to communicate to Don Lope that he intends to help him escape. He may just be a small rat but he's clever and nimble and has hundreds of brothers and sisters in the jail. Working together they'd be able to get him free one way or another and already have several plans. All Don Lope would have to do is to try and keep his nerves steady while the rats are trying to help.
Sadly in his fright Don Lope wasn't really paying attention to the rat whose Spanish if very eloquent was delivered in a rather high pitched chittering voice. Perhaps if he had listened more closely he'd have been mentally prepared for when the rat returned later that night with hundreds of his brothers and sisters to free him. If he knew their intent then he might have managed to stay calm as they swarmed around him, gnawing at the chain to release his bindings. It was just as well that they'd left his gag, though Don Lope did get rather quieter when he fainted dead away and requiring some painful nips to his tail to revive him.
