Main question is, what changed him?

Lex is a demon-vampire hybrid and I don't think those come naturally. Its a risky process if I can recall a lore page correctly, and can have side-effects or otherwise be fatal to make someone a hybrid. I'm guessing this process didn't go smoothly and lead to the events we know of.

As for who may have helped conduct the procedure? See Chapter 2, page 51 and a few pages onwards. A certain someone might not be as innocent in the situation than we have been told so far.
