It's a bit of everything for most of these, but this is just black microns and a copic brush pen for the chunky outline! I'll draw the black screen first, scan it in, and print three copies in very faint cyan (usually on the same sheet so I can see everything at once), scan and process. It's a little faster to fake it with separate layers set to multiply than to actually set up the channel separations correctly (and the result looks the same on screen - if I planned on printing these, I'd do it "correctly").
Manual color separations? Wow. If I understand your description, you are drawing each channel in black and then changing it to color after scanning?? That's just like in the old days when layout artists couldn't see their work in color until it was printed. They just had to KNOW how it was going to turn out.
Oh, and it's so much easier to work on something digitally and control the output, but it feels like cheating now. Two years ago...
Maybe not this exact piece, but I've been toying with the idea of doing something similar. I hate selling through most services though since they're far far far overpriced. On the flip side, it's hard to get large enough orders to warrant traditional screen printing :C
On the other other flip side, the shop I work at has a direct-to-garment printer (think Cafepress quality) that I can use if you really want one (and the price would be more beneficial to both parties than if I threw it on Redbubble/Cafepress) :)
