A kami this old and powerful will be very perceptive. He won't fail to notice that the volcano dragon bodyguard is very polite and knows royal etiquette. But at this stage, he's more curious than suspicious. He's hiding something, too - the nature of his "very important experiments."
Well, as they say, game recognize game. kami can recognize one another in the folklore, and with Virmir adding a sort of "kami-vision" he would be able to see Roko's spirit. That's why he immediately dropped his disguise: Kitsune can see through all illusion, so it is proper etiquette to appear to another kitsune in True Form. He started getting VERY angry when he thought Roko was dissing him, but then discovered Roko's locked into the balloon animal form... even his dispel didn't affect it, since it wasn't an illusion but physical reality. That quickly changed anger to intense curiosity.
Most of the story "Princess Saida" is telling is truth. No doubt he can see the body-swap if he chooses to, but has no reason to suspect their story at this time. Besides... he has his own agenda.

Suzocon-sama may not be fully omniscient but he can see the same as Roko, probably taken up to 11. Brand says they are body-switched, he will instantly be able to perceive the truth. You notice he tried a subtle dispel on Roko's "disguise" but found that it was not a disguise at all - he's actually a balloon animal. Which, even for a millennia-old being, has got to be completely novel.
But as I said below something is rotten about this entire arrangement. The million yen question is, what experiments is this yao no kitsune actually conducting? Notice, he did not specify that. If I were them, I would tread with extreme caution. Saida-sama and Roko-san know this instinctively. Brand? To quote Avatar, a rock Sees more. This could go very bad, very fast, if "Princess Saida" says the wrong thing to this kami.

More their experience to date. They've so far met three demigods, all of which were split off from the original deity. And all of them were corrupted. Vetra, an Air deity is a kleptocratic tyrant. Ayal, an Earth deity was bugf**k insane. And Darya, the Water deity was both ruthless and completely cuckoobananas, holding court over an army of lobotomized slaves in a crumbling "palace." They're all Skeksis.
And now we have a yao no kitsune, and worse, one associated with hi, or fire. A corrupted inari, or guardian-kami, of that power level? That is an absolutely terrifying prospect and Roko knows it.
And his servant is a Heartless meat puppet. That is a very strange thing for a guardian deity to be keeping around. And about those experiments...
In the Shinto pantheon, one can have absolutely impeccable etiquette and be completely evil. Kitsune are known for deception and misdirection. They ought to trust this kami about as far as they can throw his shrine.

As Suzocon-sama would say himself - interesting.
He is a acting as a kami of his stature should act. And "Brand", being familiar with royal protocol and etiquette, knows how to respond politely. Saida is a gaijin and would not be expected to know specific kami etiquette. Sometimes, it's the intention that counts.
Wanna bet that the first one that pops off and insults their extremely powerful host is "Princess Saida?" Suzocongami has to notice that the "Bodyguard" knows formal etiquette. How long before he notices the so-called "Princess" doesn't know beans about manners?
And what the heck is his "very important research?" Something is thoroughly rotten in this shrine. It wouldn't even surprise me if that meal they are busy enjoying was Suzocon-sama's last "experiment."

Based off of the world that he lives in, he's probably studies the effects of magical artifacts or something magically related.
We can only assume that it's something that he's been working on for knowledge since he referred to the activity as "very important research" which might imply that he's the equivalent to a scientist on this planet or possibly a philosopher???
Wait a second... now that I'm thinking about it, he might be researching on how the body behaves without a soul, Miyo might be one of Suzocongami's experiments.

My best guess, mostly based on the patterns I've seen with the other three demigods, is that he craves power. That's the one thing all of them had in common, besides being corrupt and evil - they all craved more power. Darya wanted to take over the sky. Vetra ruled with an iron fist over her domain and was constantly expanding it. Ayal was destroying his own forest to sequester all the water to... make himself more powerful.
What would make Suzocon-sama more powerful? The ninth tail. That would basically make him the Shinto equivalent of Q. My guess is these experiments of his are meant to find a way to cheat-code his way to the ninth tail and absolute power - even if he has to eat souls to do it. Miyo-san is indeed one of those experiments, but perhaps a failed one.
It's all supposition, but it's based on the comic's story thus far, and what I know of the kitsune folklore... which Virmir himself seems to have broad knowledge of.

Indeed, the semi-immortal life forms that they previously encountered are absolute megalomaniacs.
But there is the possibility that we can both be wrong about Suzocongami's intentions, only Virmir knows truth about this one, until he makes more pages in this comic.
Until then, the closest thing that we have to an answer is to speculate online, and I do have to say this was quite the enjoyable speculation.
Your knowledge of the kitsune in folklore and predictions based off of what we've previously seen has proven to be quite accurate so far, and I'm quite impressed.

I'm an old study of Japanese folklore and culture - having Japanese in-laws doesn't hurt! I call myself a weeaboo (yes, I know what that means) all the time. I'm a writer so I always try to guess characters motivations and actions. But I could be completely wrong! The fun isn't in being right, it's just finding out and having fun.
I wonder if the lie is even necessary at this point, I get that the three of them are just doing this, possibly, to build trust and avoid more conflicts with other mythical creatures, but I get the feeling that at some point someone is about to realize their true intentions :P
He mentioned he was doing some "very important research," but notably, did not specify what that research is. I suspect I know its purpose, if not its nature, but the fact he conveniently left out the details is something I would guess, say, Saida, would pick up instantly. The worry is Brand, who always is blunter than a boulder and about as subtle, and they are in the shrine of a easily offended kami with immense power.
