When a human becomes a Nemian knight they will be reborn into a superior body, gaining signifficant physical strength and size as well as enhanced senses and reflexes. However for some there is a trade off where they may lose some abilities specific to their race or culture, two examples of this are Deasia, a former Dakkaran, and Khoya, a former and only Vossen to become a Nemian knight.
Deassia once was a powerful sonomancer who had turned to a life of crime and debauchery after she went mad with power and deemed herself unstoppable. Eventually she went too far and made so many enemies that she had an army of bountyhunters and assassins after her and she found out that she was indeed stopable when she walked into a trap set by a group of bounty hunters who cut her down before she could even react. And when justice finally caught up with her that night she knew no amount of pleading, manipulating, magic or bribery could save her as she faced the consequences and every regret of her life. She was stripped of all her ill-gotten possessions and both her hands were cut off as proof of her downfall and she was left for dead in the mud. fortunately, or perhaps unfortunately the mud had sealed up most of her wounds and stopped the bleeding,saving her life for the moment to face a slow death instead. Days later she was found by a Nemian paladin whom she begged to end her suffering, instead she was taken to the monastery where they nursed her back to health as best they could and offered her a chance at redemption or properly face swift justice by Neme. Daessia wasn't sure how she could even be forgiven or what exactly redemption would mean but she decided accept the offer and go through the trials. After her ascension she found out that she could barely even use sonomancy anymore, trading that power for another form of power as one of Neme's daughters and use that power for a greater good. While she has a new pair of hands now, Neme had judged that she would forever bear the scars of her sisters' claws on her body as a reminder to never go down that path again, and the scars hurt all the more when she chastises herself every eaving according to old Dakkaran tradition.
Khoya's story is somewhat similar in that she was exhiled from the island for murdering her sister in order to gain power within her clan. As she was already an elder at the time exhile would likely mean a slow and honorless death, still she managed to survive for many years on her own but she could sense death would claim her before she could ever be allowed to return home. So she sought a warrior's death, challenging any warrior she met, but none could great her that death leading her down a path of even more senseless kiling. Untill one day she met an elderly Nemian paladin who she challeneged to a duel as well, but when she won she stayed her blade asking to be struck down instead. To her surprise the paladin offered her a place to rest and stay for a while and after some convincing Khoya relented. She stayed at the monastery for a while observing the residents in their daily chores and watching the warriors train, in many ways it reminded her of home where although there are obvious differences in ranks in society everyone still works together for a greater good and she wondered how her clan fared now without any experienced leader, all because for one moment she got selfish and how she remained selfish in looking for a warrior's death. She started to help with some chores and talking to the nuns and knights to learn more about them, occasionally accepting a request from one of the knights to spar with them and the longer she stayed and more she learned the more regrets she had that her time was running out. When she eventually learned about the ascension and was offered a chance to attempt it she took the chance at either being judged for her past life or given a chance for a new life even if she might not survive it as no Vossen or beast folk had ever attempted the ascension so it might not even work if if Neme considered her worthy.
So Khoya studied and prepared for the trials which were pretty hard on her in her old age but like any Vossen she ignored the pain and pushed on untill the day came that she could attempt the ascension. While Vossen are usually in control of their emotions she could feel the tears welling up in her eyes and her old heart pounding as she stood before the high priestes and got almost overwhelmed with emotion when the high priestess embraced her for a moment before she was led into the ritual chamber where she fell to her knees to await her fate. To everyone's surprised she emerged from the sanctum as a Nemian but she was welcomed to cheers as any new sister; To Khoya it was bitter sweet because although she got a chance to redo her life she could also never go home again now, but she had a new home and family here now and reclaim her honor serving to protect others and even her homeland in some way fighting forces that would threaten it from afar. She was allowed to keep using her old armor and weapons even though they had to be adjusted for her new size, It wouldn't make her stand out more anyways as the result of the ascension for a Vossen appear to be unique either way. Rather than having a fur color based on her natural hair color it seems she has kept a similar fur pattern and colors, down to the half white and striped tail and she also seems to have recieved a larger mane. While she doesn't feel much different aside from the vitality of youth, she has noticed some of her senses have dulled slightly while others are slightly improved. her biggest shock however was that she could now use magic, even if it was limited to Neme's sun magic, and it took her a long time to get used to using and she found that her strikes weren't quite as powerful as they should be, because this new body wasn't attuned to channeling her "Ki" effectively into her attacks. (Vossen are technically capable of magic, but they have adapted so much to channel the mana, or ki as they call it, into their attacks that they have forgotten how to use magic as intended) Though she has settled into her new life now, she has made some attempts to reach out to her old clan to find out how they are doing now, but not surprising none have responed yet.
These two examples show that the ascension can have different effects to some, and there may be a trade off in abilities lost to abilities gained, it is also strange how it effects Vossen so differently but untill more beast races join the church and become Nemians we can only guess as to how it would effect them. Would a Lipane lose all their magic as well and would they have a different pattern like Khoya? And what about lizard folk or even dragons and dragon kin, would they even undergo transformation at all given their traits and origins? We might never know for sure.

Only women are allowed to join and become Nemians, men can be given temporary shelter at the monastery but they are not allowed to join the church.
It's an ancient tradition that's the reason for this as back in the day that was the only way for women to fight in wars. And while some things changed that is the only rule that survived because the transformaion can only be performed by the sun god who lives there, and they refuse to transform males.

To answer if there are trans people in that world, Yes and no. Not in the same sense as in our real life world since medical science hasn't advanced anough to provide any surgery or things like hormone replacement. However magic exists there so people could change their biological gender that way, or illusion spells to change their outward appearance, the drawback is such magic tends to be temporary and can be dispelled. There are ofcourse curses with a more permanent effect that have been studied and reverse engineered so to speak to have a long term effect to change someone's gender. As for getting into the Nemian church, if a gender changing spell is used to decieve the order the penalty will be severe, and if a person under such a spell would pass the tests, the high priestess can detect such spells and Neme can as well, some have tried this in the past to find out the order's secrets but Neme does not tolerate such attempts at deception and none have lived to tell what they have seen. The scenario you suggested if a person is honest, then unfortunately they would still not be allowed to become a Nemian. Part of the reason is the process would not make them female anyway, so even if Neme would agree they would end up as a male Nemian. And if the person has used a changing spell, illusion or potion it would interfere with the process and could be dangerous and even deadly. There are no loopholes there to become a Nemian.
Going back to the hypothetical scenario if such an individual shows up wanting to join the church and they truly believe and feel in their heart, mind and soul that they are a woman and the high priestess can confirm that as their true self... Maybe they could be given an unofficial role with the church and serve as a cleric or squire to a paladin if the high priestess can convince Neme that their intentions are pure. They might still be limited to certain parts of the monastery however, because there are women there who are dealing with traumas from war and still healing from physical and psychological scars so there must be consideration for them hence the restrictions on movement to prevent any trouble. Perhaps they might be allowed to have a long term/permanent changing spell cast on them after a long trial period so the restriction on their movement can be lifted.
Apologies for a bit of a long winded reply, but it is a bit of a complicated and touchy subject and i'm trying to not cause any offense.
Don't get me wrong, I am kind of glad you brought this up as some things I would have to go over again if this world is ever going to become a D&D style tabletop game in order to allow some more freedom for players to create their characters, but at the same time I think there have to be some firm rules that cannot be broken, there can be a bit of give perhaps, becaus otherwise it would take away from what makes a class or race unique and make choices rather meaningless in my opinion.
Anyways, I may have to revisit this at some point to see where the rules can be bent a little. I do try to have some forms of representation with some races, the Iluurai for example where the males have breasts and can breastfeed can be considered intersex or trans, there are some types of lizard folk who change their gender at certain stages in their life, though I have not specifically mentioned them yet I think. Salamanders and gnolls are technically also intersex since their females have a pseudo-penis and Salamanders are rather androgynous looking as well since they aren't mammals. There's also a few races like the Lipane who are entirely bisexual, and same sex relationships are relatively common, though perhaps not always as accepted in some places, and there are still a whole bunch of places and races that I have yet to go into, so maybe I'll look into some alternative for the Nemians that any can join if they don't like this hypothetical scenario.

Indeed, I only brought it up on this piece because it was the most recent and figured spesifically female related content, and i brought it up solely because I was curious if the idea had crossed your mind before. As you said, if you're ever going to make it into a TTRPG, you need answers for this sort of thing.
I've certainly not been offendeded by anything you've posted here and very much look forward to learning more about the setting, maybe even playing it one day.
