One layer for the background, and one for the wolf. I hardly use any photoshop specific technique: just a round brush with pressure on size and 50% fixed opacity, 70% for the preliminary blocking of shapes. Sometimes i use layers for corrections and effects (like a multiply layer for adding the tattoo), but i flatten them almost immediately.
Another technique i use is to duplicate a layer, and do colorize/brightening/darkening the copied layer, then create a fully transparent layer mask and painting over the original using the modified version.
In this picture i created a duplicate wolf, turned its color into bright red, then merged the original and the modified version via layer mask painting. Then flattened the 2 layers to have a definitive version of the wolf. That's something i use very often too.
