Pfff it's the primers + MasterMix
I actually never put them on ice, since the support comes directly from the freezer into the cold PCR room, the reagents are always stable. I got the microsatellites to work in the end, put it did took me all Summer to optimize them. It was mostly 454 primers not being specific enough for the protocol we wanted to use.
