Hey spinningpeanut! Actually there are no tweens like what you might use in Flash - Lance was referring to the inbetweening process, which was done by hand.

I use Plastic Animation Paper ( free program: http://www.plasticanimationpaper.dk ) for the rough animation, and Toon Boom Animate Pro for inks and color.

Well Plastic Animation Paper is easy once you get the hang of it, but hard to learn. Also it has some quirks you have to work around.
Toon Boom is a big program and takes a while to learn so I guess that's the harder one. But it doesn't play tricks on you like PaP, although it can crash from time to time if your comp runs out of memory.
