Manga Studio 5 is a great program. But the tool naming/translation from Japanese leaves a lot to be desired.
Here's a quick tutorial on how to create comic panels. Which, I admit took me a while to figure out. It's still easier than MS4 Debut. I understand that MS4 Ex is quite a bit easier than Debut, but I'm cheap. :)

I love Manga Studio. But honestly, I find that the "panel" tools just make things more complicated than they need to be. I usually just pick some appropriate rulers and draw the panels in myself.
........ course, I haven't done many comics lately, so take my thoughts with a grain of salt.
