Hummm, Rebecca "Capricorn" in her normal scrawny teenage goat form seems to be the only 'appropriate' match-up for the teenage Bengal tiger Gabriel "Cancer" in an actual physical fight. However if Gabriel gets to compete in a video game with another team member (due to his being underage) that might be a whole different story...
According to their Bio Sheets, the twelve Zodiac team member's ages are:

There are some interesting issues with how to setup something like this, especially in the superhero genre where character abilities can vary by orders of magnitude.
Without some sort of 'power inhibitors' the characters with super durability and super strength have such a large advantage in a "strictly hand-to-hand combat" sparring match without weapons or powers that they are in a different league than those whose physical strength and durability are essentially the same as 'normal people'. As Gemini (who may be the best hand-to-hand fighter on the team who doesn't have superpowers) thinks to himself when facing off against the mind-controlled Aries, "He's like a tank. I don't think I could stop him if I wanted to." Similarly, among the characters whose physical abilities are within the range of 'normal people' there will be significant advantages to those who are large/strong and those who have hand-to-hand combat training and experience. I think Ramas has indicated that some consideration has been given to those issues with his comment that, "the brackets have been determined by myself and the other directors." Thus, like many sporting contests, the winners may be determined as much by the rules of the contest and how the matches, 'brackets' or leagues are determined as by the actual ability of the participants.

Well there are a dozen Zodiac team members so that would be six one-on-one matches if everyone were to participate. Here's one possible way to set up the matches:
1) Gabriel "Cancer" vs. Rebecca "Capricorn" (the short scrawny teenagers)
2) Sonja "Scorpio" vs. James "Libra" (roughly similar size and age, Virgo could also be a potential match)
3) Kyra "Sagittarius" vs. "June" "Aquarius" (trained, experienced professional women... and Kyra vs. Pete seems like a bad idea)
4) Luna "Virgo" vs. Pete "Gemini" (not a 'good' match, but Virgo likely needs to learn something about mismatched fights and dirty fighting)
5) Janine "Taurus" vs. Kenneth "Pisces" (Leo or Aries would be other matchups for Taurus, otherwise either Gemini or Pisces, and Taurus may need to learn how to be brutal enough to win)
6) Wade "Leo" vs. Kelly "Aries" (it is difficult to find an appropriate match for Aries, and according to Morris, Ramas likes, "likes to see you frustrated, Mr. Leo")
Of course, Pete "Gemini" vs. Kenneth "Pisces" would be an appropriate match and Pisces even commented that this is a good match up, but the need to use all twelve members only once led me to the matches above. Since Ramas seems to have some prophetic insight and be more than a bit eccentric, the matches could vary wildly or have unexpected additional rules or conditions.
Does anybody else have a suggested set of one-on-one matches that use all twelve Zodiac team members only once each? This could be a fun thing to speculate about and see other people's thoughts and ideas.

One way to do additional rounds after the first set of six one-one-one matches would be to eliminate the loser of the lowest numbered match and the winner of the highest numbered match and then for all ten of the remaining team members the winner of each match takes on the loser of the next higher numbered match, for five additional fights. Although it might get tedious to draw (and thus be subject to some cut scenes), this process could be continued with eight, then six, then four, and finally two (for positions #6 and #7 of 12), leading to a final ordered 'ranking' of the hand-to-hand combat ability (in a sparring match held in a ring) of all twelve Zodiac members from lowest to highest.
There is probably actually at least some value to the team leader(s) like Leo and the executives of knowing the team members' current respective rankings in hand-to-hand combat, and some of the matches are likely to be learning experiences for the participants (and possibly the spectators) as well as show what some of the team members need to work on, so this 'tournament' might not be quite as much of waste of time or as eccentric/bizarre as it initially appears to Leo and Morris.
From a sporting or drama perspective, this scheme maximizes the number of more competitive matches where the outcome is actually in doubt while minimizing the amount of ridiculous mismatches. Note that with this scheme and the original set of matches above it is unlikely that there would be a Kyra vs. Pete match before the third set of matches. It is possible that by the third set of matches there might be rematches of the original pair which could lead to a different result than the first match between that pair since they would be fairly evenly matched. It is even possible that Gemini's second and fourth (or fifth) matches could both be against Pisces (with Gemini winning the first one and losing the second time) and ultimately resulting in the final match (for positions #6 and #7 of 12) being Pete vs. Kyra, which might be the ultimate in drama (not that Ramas might have planned it like that or anything).

You have thought about this a lot my friend and have some very good guesses. The match-ups were not determined by me but by the commission customer. I will tell you this. The plan is to have a guys vs. girls match up so each one will be one of the men vs one of the women. One of your predictions is correct.
We just found out that Round 1 is Taurus vs. Aries: https://www.furaffinity.net/view/56887503/ so I decided to update my thoughts on matches.
With six female and six male Zodiac team members, here's my guess on one possible way to set up the matches the six one-on-one female-vs-male matches:
1) Rebecca "Capricorn" vs. Gabriel "Cancer" (inexperienced, short scrawny teenagers)
2) Luna "Virgo" vs. James "Libra" (inexperienced, roughly similar size and age)
3) Sonja "Scorpio" vs. Kenneth "Pisces" (experienced)
4) Kyra "Sagittarius" vs. Pete "Gemini" (experienced, I can imagine a commissioner putting these two up against each other)
5) "June" "Aquarius" vs. Wade "Leo" (Leo and Aquarius is a bit like Sag vs Gemini)
6) Janine "Taurus" vs. Kelly "Aries" (the obvious male/female brick pair-up)
